Abstract

The thesis investigated blood specimens to examine hemoglobin types of 20 Sagai from Palian District, Trang Province and 76 Chong from Tambon Khlong Phlu, Makham District, Chanthaburi Province. The investigation revealed that the Sagai originated in the Southeast Asian region. The Chong and the Khmer originated from a minority Khmer group or from cross-marriage with the Thai. It was hypothesized that the Chong might originate from two sources (pp. 88-89).
    
The study results from investigating blood specimens of the two ethnic groups were divided into three parts. Firstly, the hemoglobin examination of the Chong through Cellulose Acetate Electrophoresis and measured by Microcolumn Chromatography revealed that 14 Chong (18.4%) had the hemoglobin type A2A at the volume of 2.08-3.3, that 33 (43.4%) had the EA hemoglobin at the volume of 25.7-35.4, and that 29 (38.2%) had the EE hemoglobin at the volume of 75.3-83.3. As for the Sagai, 19 (95%) had the A2A hemoglobin at the volume of 2.13-3.43, and only one had the EA 30.24 type (p. 64). Secondly, the examination of the HbE gene by means of the ASPCR method to discover the Beta E-globin was conducted with the Chong of three hemoglobin types (A2A, EA and EE). They were randomly selected for the numbers of 5, 21 and 17 from each group respectively. It was found that 400 bp DNA on agarose gel (12%) was found in those with EA and EE types, but was not found in those with the A2A type. When it was examined for the Beta E-globin, 400 bp DNA was found on the electrophoresis gel (p.65). Thirdly, from examining the Haplotype characteristics within the Beta-globin genes, it was revealed that Sagai genes contained Beta E1 chromosomes with Haplotype as heterozygote. It could be confirmed that the origin of the Beta E genes of the Sagai originated in Southeast Asia. For the Chong, their Beta E genes were mostly found on FW3 microsomes.
     
From this investigation, it was indicated that the Beta E genes of the Chong and the Khmer were from the same origin. A small number of the genes were found on FW2 chromosomes. It was believed that their Beta E genes were from a minority Khmer group or from cross-marriage with the Thai (p. 88). It was concluded from this experiment that the Beta E-globin genes of the Chong originated from at least two sources (p. 89).
